genomeCoverageBed -ibam SRR2048235.bam -bg -trackline -split -g mm10.chrom.sizes > mytrack.bedGraph
bedGraphToBigWig mytrack.bedGraph mm10.chrom.sizes out.bw

Those are the processes I used to generate the bigwig file out.bw. After I pasted the url to UCSC, I got this error message:"http://ch8316f5eyu.220idc.cn/test/out.bw is not a bpt b-plus tree index file". Does anyone have the idea what is going on?

The file became corrupt either during creation or (more likely) during upload. For example there's an extra byte that's inserted at position 6 (value 13) that's shifting the remaining bytes by 1. Reupload the file and see if that solves the problem (if not, delete it and make it again).

Actually I simplify this problem. At first I wanted use Homer to generate multiwig to overlay tracks in UCSC, and I got this problem after paste url in track hub. In a word, I have tried so many times and this problem always occurs............

Your response serves no purpose. I have already told you (1) why this is happening and (2) possible solutions. The fact that you used Homer is completely irrelevant.

Thanks. You are right. The file worked fine for IGV before uploading, but became damaged when I download this file from this url and did not work for IGV. It is weird that uploading can damage file every time........
