Larry's comment highlights an important point. Your question is very very basic, akin to asking Yo-Yo Ma, "How do I make music?" It would be much better if your question had more detail, both to set the context and offer people more hooks into how to answer it, but also to give people more of a reason to read it, to see what you're doing, to see if one path is better than another, to see if someone has already done something similar to what you're trying to do. You'll get much better answers, and your question itself will be informative to some. Things like: what organism? How many ESTs, average length? Library characteristics? etc.

Try BLASTing them against organisms about which something is known,and you'll be functionally annotating them.

It is also important to use the information about the library/libraries from which the cDNAs were generated. This can give you data on tissue/cell type where teh gene is expressed, under what conditions like treatment or developmental time point. While direct aspects of functional annotation, putting that annotation into context of tissue, developmental time point and such is useful.
