My personal suggestion towards a must-read for scRNA-seq would be the extensive and comprehensive http://bioconductor.org/books/release/OSCA/. This book contains example code, explanations and discussions plus hands-on guidance using several example datasets which can be easily loaded via the scRNAseq
package without the need to do any low-level processing. From this on you can dive into the vignettes of the suggested tools and start building an analysis pipeline suiting your needs.
