Jay Flatley’s announcement yesterday certainly changes calculations for whole genome sequencing. Newer, cheaper optics, fluidics and reagent chemistry have lowered the cost of sequencing and enabled a 300 cycle, 125 Gb run in 30 hours with the NextSeq 500. The HiSeq X Ten, (pronounced ex ten, not ten ten) consisting of 10 instruments daisy chained together, will generate 18 Tb in 72 hours. The new optical technology now utilizes a 2 dye system: adenine and cytosine bases are represented by one dye, an absence of dye for guanine bases and both dyes for thymine. For anyone interested in reading further, there have been at least three articles on GenomeWeb this week describing these announcements (link to the first announcement here). The big catch line about the HiSeq X Ten,

He (Illumina President and CEO Jay Flatley) also said that the firm's technology can now produce a human genome for under $1,000, claiming to be the first to achieve this long sought-after goal.

I was very intrigued by this statement at first but I don't think this is going to be a relevant figure for most researchers. The NextSeq 500 is said to produce "HiSeq throughput in a MiSeq-sized box" and I think this instrument is aimed at research use. The HiSeq X Ten is only sold in units of 10 and costs $10 million (as opposed to $250k for the NextSeq 500), and is said to produce 5 genomes per day (as opposed to 1 genome/day with a HiSeq). Given that this announcement happened at the JP Morgan Conference, combined with the price and a throughput of ~50 genomes/day, I think we can assume this instrument is targeted towards industrial-scale use. I can't imagine many research institutions having this kind of demand but it seems prime for clinical applications.

The statement "Ten Illumina sequencers daisy chained together." really got me thinking. It reminds me of those ad-hoc "wiring solution" where one has a bunch of extension cords plugged into one another.

The HiSeq X Ten is also limited to human whole genome sequencing. I suspect that these limitations (10 at a time and WGS only) will be lifted in 2015.

I've read this elsewhere, but the NextSeq 500 looks very much like something to try to steal market share away from the IonTorrent.
