minor peak in TSS enrichment plot

Hi I am using signac for single cell ATAC-seq analysis.

I have a question about TSS enrichment plot, one of the QC plots:

What does the minor peak to the right of the major peak stand for? Does it have a biological meaning?

Hi linzhang stefanie,

I would like to know if you find any solution to your doubt because I have the same one and the only thing I found was that It is the spacing region between two flanking nucleosomes but I am not 100% sure of that reason.

Yes, isn't that second peak just the expected biology as you say based on the nucleosome positioning? You see this also in published plots all the time. Nothing to worry about if you ask me.
