Recommendations for API/Flat file DB describing physical and biological properties of human proteins

I am looking for an online database of the physical and biological properties of all/as many human proteins. Recommendations are appreciated.

The two main areas of interest are:

1) The properties of interest of the protein include domain (ECM, inter-cellular, nucleus), membrane-bound/spanning, hydrophobicity, homo-oligomeric, hetero-oligomeric, transition metal-containing, co-factor containing, etc.

2) Some measures of biological activity between a drug and a protein.

Ideally, if there is an API I can connect to, that would be great.

Have you looked at UniProt? https://www.uniprot.org

Human proteome: https://www.uniprot.org/help/human_proteome

Subcellular location, transmembrane region, subunit structure, binding sites (including metal), cofactor etc are only some of the concepts that are curated in UniProtKB/Swiss-Prot.
