. In any case, if someone could point me how to know this from the file itself, that would also be great.

Looks like someone asked the developers about this indirectly last year but the question was never answered: https://github.com/marbl/Mash/issues/177
If you knew the version of RefSeq used to build the file then you can probably look up the number of sequences from RefSeq release notes.
