Hi, I have a doubt regarding the use of Illumina Dna Prep kit (or other kit based on tagmentation/transposons) for the sequencing of linear genomes, specifically phage genomes that in some cases have specific termini (genome ends). In some articles i have read that they cant't be used because a transposon can't insert upstream the genome termini (because there are no more nucleotides) so these sequences will be lost. How can this kind of kits grant the sequencing of the very ends of linear genomes (or even telomeres in human chromosomes)?
Thanks
