I am new to single cell analysis. Can RNA velocity (spliced/unspliced) be calculated solely from the raw file (count matrix) provided in the Supplementary files from GEO?

First question.

1. The raw file (count matrix) provided in the Supplementary files from GEO would be spliced data, right?

Second question.

2. Can RNA velocity (spliced/unspliced) be calculated solely from the raw file (count matrix) provided in the Supplementary files from GEO?

Thanks

No, you either have the spliced and unspliced counts available that you need, or you need the fastq files to generate them. Before you dive into analysis or you should read this:

https://www.sc-best-practices.org/preamble.html

It will cover many of the necessary basics you need to know before really getting into the "beautiful" scRNA-seq world.
